Guidelines for DNA Sample Submission print this page   email this page


We understand that every sequencing result and turnaround time is critically important for you to proceed to the next step of your research. Please review the following information for details on how to submit DNA samples to ACGT, Inc.

 Full Sequencing Analysis

- We accept purified plasmid, bacterial clones, PCR   DNA, purified or unpurified, BAC DNA for analysis.

- We require plasmid or PCR DNA as follows:

PCR DNA 500 ng per kb > 20 ng/µl

- Reference sequences should be provided in FASTA   format.

- Please include a photograph of the template DNA   with the quantity of the DNA loaded and a molecular   marker with the order.

 Bacteria with BAC or Cosmid for Large Insert   Sequencing

- Sponsor should summit at least 100 µl of glycerol   stock grown in LB media with 10% glycerol for large   insert sequencing.

- Although different types of containers are accepted   for sample submission, 1.5 ml or 650 µl microfuge   tube is recommended. Glycerol stocks should be   submitted via frozen package.

- Reference sequence for each sample, if available,   should be provided in FASTA format.
   Purified BAC or Cosmid for Large Insert   Sequencing

- Each sample should contain at least 50 µg of purified   DNA, preferably processed using QIAGEN Large   Construct kit.

- DNA templates should be of high molecular weight.   Sample can be submitted as a pellet or resuspended   in water or alcohol.

  DNA Samples For Shotgun Sequencing

- Sponsor should submit at least 200µg of purified   DNA template for shotgun sequencing.

- DNA templates should be of high molecular weight.   Sample can be submitted as a pellet or resuspended   in water or alcohol.

  DNA Samples For Shotgun Sequencing

- Sponsor should submit all Test Articles (PCR   products, plasmids, cells, etc.) for GLP-compliant   protocol studies in frozen package.

- Information on Sample ID, sample collection   documentation, and packaging information should be   submitted along with samples.

- Upon receipt, the integrity of sample containers will   be checked by analysts and relevant information will   be recorded on sample log-in record or laboratory   notebook.
