General Guidelines for Quick Single Pass DNA Sequencing Service Samples print this page   email this page


We understand that every sequencing result and turnaround time is critically important for you to proceed to the next step of your research. In order to generate good sequencing data the first time, it is essential to consider the following guidelines when submitting DNA samples.

 Template DNA Information

1. Indicate the size of plasmid DNA on the order form. For large DNA, we use different cycling conditions and longer extension time.

2. Indicate the size of the PCR DNA on the order form. We adjust the template amont for sequencing, depending on the size of the PCR product.

3. Accurately indicate the DNA concentration. The DNA sample quality and quantity are randomly checked by Nanodrop or agarose gel electrophoresis.

4. If your DNA concentration is very low or your sample is suspected to be dirty, please let us know in advance and send us extra DNA. We will be happy to modify our sequencing reaction conditions to produce clean data the first time.

5. For sequencing the PCR product, DNA concentration needs to be cheched after amplification, and again after clean up. We can also clean your PCR DNA prior to DNA sequencing.

6. Indicate if your DNA sequences contain high GC, repeated sequences, or poly A, so we can modify sequencing reation conditions.

7. Indicate if your DNA was purified by CsCI or large scale purification.

8. We honor your special request, such as longer sequences, closer to priming site, or short sequences. Please indicate your request, so we can meet your needs at the first time.
   Sample Name Clarification

1. The name of the sample on the tube and on the order form should be identical.

2. The name of the template DNA should be different in every order. Do not use the same template name more than once.

3. Do not make your sample name too long. Any combined template and primer names with more than 28 characters will be truncated.

4. The only symbols accepted by our sample naming system are underscores(_) and dash(-). Please avoid using any other symbols or space in your primer and template names.

5. If you use tubes with screw-on-caps, please label both the tube and the cap.

  Shipping

1. Use proper protection for your shipment to prevent sample tubes from braking. We will try our best to recover samples from cracked tubes. However, DNA may be contaminated and may not generate clean data.

2. To save shipping charge, try to consolidate orders withing your group rather than sending in several packages. However, each order form should match your DNA samples in one bubble bag.

3. When shipping DNA samples for overnight delivery, dry ice is not necessary unless your shipping policy requires it. Room temperature shipping also reduces your shipping cost.
